众所周知,生物钟与生命活动密切相关,它控制着几乎所有包括我们人类在内许多动植物的生物过程,包括我们的睡眠-醒觉、动物的昼行夜伏、植物的春华秋实,以及我们的代谢功能和认知过程。

此前有研究表明,这种以24小时为周期的内部时钟在光合细菌中也得到证实。光合细菌利用光线产生化学能;但非光合细菌是否也受昼夜节律的支配科学家对此还知之甚少。

近日,发表在《Science Advances》上的一项研究中,来自德国慕尼黑大学领导的国际研究团队首次发现,非光合细菌中也有生物钟。它们通过光线或环境温度感知周期,使其分子适应一天中的时间。这一发现将给生物医学、生物农业和工业生物技术领域带来有价值的信息。掌握了细菌的昼夜节律,在未来,临床给药的时间将变得更加精准

在这项新研究中,研究人员选择了非光合枯草芽孢杆菌(Bacillus subtilis)作为实验模型,这是一种耐寒且经过充分研究的多功能微生物,分布在土壤及腐败的有机物中,也包括人类在内的许多动物胃肠道中。枯草芽孢杆菌已广泛应用于生物医药、动物饲料生产、植物抗病、水产养殖和水质净化诸多领域。

虽然枯草芽孢杆菌不进行光合作用,但由于光感受器的存在使它们对光线也十分敏感。先前对枯草芽孢杆菌的观察提供了线索,它们的基因活性和生物膜形成过程可能会根据环境线索(光照水平或温度变化)遵循昼夜周期。

为了深入研究,研究人员将枯草芽孢杆菌暴露在连续12小时光照和12小时黑暗的环境中,然后检测其基因表达活性。

在光/暗交替周期中,编码蓝光受体ytvA基因的表达在黑暗时增加,在明亮时减少,这表明昼夜节律在这一过程中发挥作用。

研究人员发现,在持续黑暗的条件下,枯草芽孢杆菌的生物钟仍然存在,但周期延长。在没有光信号的情况下,其生物钟没有严格遵循24小时的周期。

在另一项实验中,研究人员采用温度循环的方式观察。

同样,当温度在25.5°C和28.5°C之间以12小时为周期循环时,ytvA基因的表达也出现起伏,与光照一样,在自由运行的实验中(与环境线索不同步)循环持续时间更长。

这两项实验结果告诉我们,非光合细菌也具有生物钟。

研究通讯作者、慕尼黑大学的时间生物学家Martha Merrow说:“虽然这些发现目前只涉及一种细菌,但这是研究人员首次在非光合细菌中发现这种现象,这可能让我们对细菌这个整体的理解有着巨大影响,因为细菌占地球总生物量大约15%。”

研究人员推测生物钟可能在某种程度上受到转录-转译反馈系统的调节,也可能与代谢周期有关。目前还不清楚是否存在某种形式的核心时钟可以控制枯草芽孢杆菌的昼夜节律,就像人体中的一样,但研究人员指出这是一种可能性。

研究人员表示,研究枯草芽孢杆菌的核心时钟,或者它们是否存在多个时间振荡器将为我们提供更多有价值的信息。

无论如何,24小时生物钟对细菌的影响可能具有重要的意义,这不仅是在科学上对细菌生物学的理解,而且还包括生物医学、生物农业以及工业生物技术等领域的潜在应用。

研究共同通讯作者、丹麦科技大学的生物工程部细菌相互作用和进化组Ákos Kovács博士说:“枯草芽孢杆菌除了用于人类和动物的益生菌外,还用于日化生产,乃至到作物保护等各种领域。因此,在这种细菌中设计生物钟将使不同的生物技术领域实现突破。”
