PNAS:干细胞治疗可缓解眼压降低青光眼风险

研究人员将新生细胞注射到青光眼小鼠模型的眼内,发现新细胞的流入能够帮助小梁网状结构再生,该结构是眼部的一个排水管道能够帮助避免出现液体累积,如果液体在眼睛里累积会使眼压升高容易导致青光眼。这种疾病会损伤视神经,可能导致失明。
 
"我们认为用健康细胞替代损伤或缺失的小梁网状细胞能够重新恢复小梁网状结构的功能。"文章作者Kuehn在他们实验室的网站上这样写道。
 
相关研究结果发表在国际学术期刊PNAS上。
 
这种方法的一个潜在优势就是利用病人自身皮肤细胞获得诱导多能干细胞,这样就能避开使用胎儿干细胞存在的伦理问题,还会降低病人机体对移植细胞的排斥反应。
 
Kuehn等人利用一种基于人类小梁网状细胞培养条件开发的条件培养基成功诱导干细胞生长为类似小梁网状细胞。随后他们将获得的新生细胞注射到小鼠眼部,发现这还会导致内源性小梁网状细胞的增殖。换句话说,注射进去的新细胞不仅能够自身存活,还可以促进机体产生更多自身细胞,从而进一步提高了治疗效果。研究人员在移植9周后检测了小鼠的治疗效果,实验室小鼠一般存活两到三年,9周大约相当于人类的五到六年。
 
研究人员表示目前青光眼还无法治愈,只能通过控制病情防止失明。目前主要的治疗方法就是使用滴眼液,进行激光或者传统手术。他们认为这项研究已经带来了常见青光眼治疗(原发性开角型青光眼)的希望。
