Biomaterials:载有奥替普拉的PLGA纳米粒子可有效治疗急性肾损伤和纤维化

为了应对这一挑战,我们在此证明100nm直径的聚(乳酸 -共-羟基乙酸)(PLGA)纳米颗粒(NPs)可以选择性地在小鼠损伤肾脏中累积,与肾缺血-再灌注损伤的初始阶段的肾损伤程度和给药时间相关。NPs位于通过免疫荧光证实的肾小管上皮细胞中,这对于AKI的进展是至关重要的。利用PLGA NPs在缺血再灌注(IR)肾脏中的高积累和肾小管靶向,我们设计了载有Oltipraz(PLGA-Oltipraz NPs)的PLGA NPs来治疗IR诱导的AKI和肾纤维化。

体外实验结果表明,与游离的奥替普拉相比,PLGA-Oltipraz NPs具有更高的抗氧化作用,提高了细胞活力,降低了丙二醛含量,提高了超氧化物歧化酶的活性。PLGA-Oltipraz NPs的治疗功效在体内进一步研究。PLGA-Oltipraz NPs处理的AKI小鼠在初始阶段显示出明显减少的肾小管坏死,较少的胶原沉积和更好的肾功能以及在恢复期改善的肾纤维化。

总之,该研究为PLGA-Oltipraz NPs治疗AKI和纤维化提供了一种有前景的方法。它还揭示了尺寸选择性NP和药物施用时间窗对纳米疗法的重要性。
