Plos One:抑癌基因p53降低肝癌上皮间质转化,抑制转移

肝细胞癌是癌症致死率最高的三大恶性肿瘤之一,其高度转移的特性成为临床治疗的最大障碍。肝癌病人的癌组织中常见p53基因的突变,尽管p53在肿瘤起始及发展中的生理功能已经得到详尽的阐述,但p53在肝癌细胞转移中的作用仍有待研究。

在研究员陈雁的指导下,博士生王征等探讨了p53在肝癌细胞EMT及转移中的潜在功能,发现insulin和TGF-beta诱导的EMT以及细胞迁移能力均在敲减p53后进一步加剧。此外,研究发现p53通过影响beta-catenin信号通路可发挥对EMT的调控作用,降低p53表达能够促进beta-catenin的细胞核内积累及转录活性,抑制beta-catenin信号通路可减弱p53降低所促进的肝癌细胞EMT、细胞迁移和肿瘤转移程度。该研究揭示出,p53通过调控beta-catenin信号通路可对肝癌细胞EMT及转移发挥重要的功能。

据悉,陈雁研究组在前期的研究工作中发现,p53参与了皮肤肿瘤形成过程中的EMT进程,并提出p53是一个调控EMT的“关卡”。
