在微生物中,进化上保守的机制通过应激诱导突变(SIM)可以促进其对恶劣条件的适应。类似的过程可能是人类癌症进展和治疗失败的基础。

最近,研究人员描述了在非遗传毒性药物选择下,人类癌症的多种体外和体内模型中的SIM,其反常地以牺牲内在健康成本来增强适应性。

一个全基因组的方法确定了雷帕霉素的机械靶点(MTOR)作为一个压力感应器,介导多个癌症类型和条件下的SIM。

这些观察结果与耐药性的两阶段模型相一致,其中遗传多样性最初的快速扩张被内在的适应性惩罚所抵消,随后在新的条件下正常化到完全适应。

这个模型提出了合成致死策略,以最大限度地减少对抗癌治疗的抗药性。
