最近发表在《细胞》(Cell)杂志上的一项新的研究描述了一种糖尿病相关基因对一条生物信号通路起作用,影响胰岛素释放的一些细节。研究作者称,找到一些能够对这一信号通路起作用的药物,或许最终可促成针对1型糖尿病的新疗法。
研究的共同领导者、费城儿童医院应用基因组学中心(CHOP)主任Hakon Hakonarson 博士说:“2007年,我们的基因组学研究小组在一项全基因组研究中发现了第一个在1型糖尿病中起重要作用的基因,但我们不知道它的功能。现在我们了解了这一基因在调控胰岛素代谢中发挥至关重要作用的机制。”
宾夕法尼亚大学Perelman医学院糖尿病、肥胖与代谢研究所的Doris A. Stoffers博士与Hakonarson是这项研究的共同资深作者,也是通讯作者。
当前的研究发现是建立在Hakonarson和CHOP的同事们2007年的一项全基因组关联研究(GWAS)基础上,当时他们证实一种叫做KIAA0305(也叫做CLEC16A)的基因变异,与1型糖尿病和其他自身免疫性疾病风险增高有关。
随后Hakonarson研究小组开发了一种Clec16a基因失活的小鼠品系。他们与内分泌学专家Stoffers合作,饲养了一组只有胰腺细胞受累的基因敲除小鼠。
科学家们发现,Clec16a基因作用于对胰岛素分泌至关重要的一条信号通路。正常情况下Clec16a帮助保护细胞的微小能量生成元件——线粒体。当Clec16a基因敲除时,一个叫做线粒体自噬(mitophagy)的过程受到影响,异常的线粒体生成较少的ATP,能量输出受损破坏了胰腺中分泌胰岛素的β细胞。“Clec16a缺失最终造成了不健康的线粒体累积,使得β细胞减少分泌胰岛素,”Stoffers说。
在人类,无法生成胰岛素是1型糖尿病的一个标志。研究小组发现,携带CLEC16A单碱基变异的人β细胞功能下降,尽管没有基因敲除小鼠中那么极端。
研究人员证实,Clec16a生物信号通路对下游的一种Parkin蛋白产生了影响,众所周知Parkin是线粒体自噬的一种主要调控因子。当前的研究第一次将Clec16a信号通路与Parkin介导的线粒体自噬调控联系起来,表明了这一过程通过导致胰岛素分泌失调影响糖尿病的机制。
如果能够开发出一些药物对Clec16a信号通路起作用,它们有可能能够为携带CLEC16A基因风险变异的1型糖尿病患者提供新的靶向性治疗。
Hakonarson说,人们常常批评GWAS不能鉴别出对复杂疾病风险造成重大影响的基因变异,但在这一案例中,7年前GWAS鉴别出的一种过去未确定特征的基因,被证实不仅在1型糖尿病中,也在多发性硬化症、风湿性关节炎、克罗恩病和其他自身免疫性疾病中起至关重要的作用。他补充说:“还需要开展更多的工作还鉴别出1型糖尿病其他的致病遗传变异。”
