常规的化学治疗方法在治疗脊索瘤时疗效有限。CLIN CANCER RES近期发表了一篇文章，研究SOX9在脊索瘤中发挥的功能及其治疗应用前景。作者利用免疫组化分析了50例脊索瘤标本SOX9表达。使用western blot和免疫组化分析脊索瘤细胞系中SOX9表达。利用SOX9siRNA抑制SOX9表达。抑制SOX9后表达后通过MTT法检测细胞增殖能力和细胞毒性情况。通过划痕实验及Transwell实验评估SOX9敲除对脊索瘤细胞运动的影响。通过western blot及流式细胞学检测SOX9敲除诱导的细胞凋亡，细胞周期捕获及肿瘤干细胞分子标志表达下调情况。使用MTT法检测SOX9 siRNA联合化疗药阿霉素+顺铂对脊索瘤细胞的影响。组织芯片和免疫组化分析表明SOX9在脊索瘤广泛表达并且高SOX9表达与不良预后有关。RNA干扰敲除SOX9可以抑制脊索瘤细胞生长，降低细胞运动能力并诱导细胞凋亡。此外，SOX9抑制联合化疗药物对脊索瘤细胞有较强的抗肿瘤作用。文章最后认为，SOX9在脊索瘤中起到了关键作用。靶向SOX9为脊索瘤的治疗提供了新的方向。原始出处：Hua Chen,Cassandra
