由IFN-α、IFN-β、IFN-ε、IFN-κ和IFN-ω组成的I型干扰素是一种有效的多效性细胞因子,具有多样的免疫功能。目前的研究也报道了ISGs的表达升高与SLE疾病发生发展相关。
ISGs编码的几种基因产物与抗病毒防御机制和细胞功能的调节相关,然而,这些基因在SLE中的生物学作用仍知之甚少。
SLE患者和健康对照的CD8+T和CD4+T细胞的转录组分析
在该研究中,研究人员通过分析CD4+和CD8+T细胞的转录组,发现IFN高表达的患者中线粒体衍生基因和线粒体相关代谢通路被下调。这些患者的CD8+T细胞中的线粒体增大,细胞的备用呼吸能力(SRC,spare respiratory capacity)降低,在接受TCR刺激后,细胞死亡增加。而将健康人的CD8+T细胞暴露于I型IFN和TCR刺激也会引起上述的线粒体异常。

机制研究显示,这些“ SLE样”状态会增加CD8+T细胞中NAD+的消耗,从而导致线粒体呼吸功能的降低以及细胞活力的降低,两者均可通过补充NAD+来进行恢复。
