放线菌素D纳米粒子治疗急性髓性白血病

Act D是一种抗生素药物,最初于1964年在美国被批准用于治疗传染病。随后,该抗生素在1974年被证明具有抗癌活性。从那以后,该药物已被用于治疗各种类型的癌症,包括横纹肌肉瘤、尤因肉瘤、滋养细胞肿瘤和睾丸癌。该药物在世界卫生组织的基本药物目录中被列为卫生系统中所需的最有效药物之一。Falini等人曾在《新英格兰医学杂志》上报道,Act D的静脉内给药可用于治疗携带NPM1基因突变的急性髓性白血病(AML)。最近的研究还表明,Act D是RNA合成的有效抑制剂,可能具有治疗COVID-19患者的潜力。

然而,该药物是通过静脉内(IV)给药,可能会在给药后的最初几个小时内由于血液中Cmax过高而产生严重的毒性。Act D纳米颗粒制剂可以使Act D的释放缓慢并且可以将血液中的Cmax预先调节至所需水平。
