CLIN CANCER RES:巨噬细胞通过改变VEGFR表达促进抗VEGF治疗耐药

作者建立了抗VEGF治疗获得性耐药的模型来研究免疫细胞在VEGF阻滞获得性耐药发展过程中所扮演的角色。在体内和体外研究中探索巨噬细胞在耐药中所发挥的作用。使用抗VEGF抗体获得性耐药鼠肿瘤模型,作者发现了巨噬细胞在这类耐药中具有之前未知的作用。巨噬细胞会招募到肿瘤微环境中且与抗VEGF抗体获得性耐药发生有关。清除耐药后出现的巨噬细胞可以抑制肿瘤生长并延长荷瘤小鼠的生存。在巨噬细胞缺陷鼠模型中,抗VEGF抗体获得性耐药无法出现,但是注射巨噬细胞后可以诱导产生抗VEGF抗体获得性耐药。下调巨噬细胞VEGFR-1和VEGFR-3表达会伴随血管生成通路上调,有助于抗VEGF治疗逃逸。

文章最后认为,这些发现为目前的抗血管生成治疗疗效不显着的原因提供了新的理解,为使用联合疗法治疗卵巢癌及其他肿瘤的提供了新的机会。
