PNAS:细菌感染之光动力抗菌新进展

细菌感染一直威胁着人类健康。随着近年来细菌耐药性严重化,细菌感染将可能带来比癌症更严重的全球性危害。光动力抗菌是一种结合光敏剂、氧气、光产生活性氧,从而破坏菌膜中活性物质的抗菌方法,它不易使细菌产生耐药性,且时空可控性高、入侵性小,因而在抗菌领域受到了广泛关注。然而,在光动力抗菌过程中,活性氧成分寿命短、作用半径小,且菌膜可有效阻挡外来物质,因而如何提高光敏剂的膜插入能力成为光动力抗菌的难点和关键

近期,中科院理化技术研究所田野团队联合上海交通大学颜徐州课题组、美国犹他大学Peter J. Stang课题组,将修饰细胞穿膜肽TAT的烟草花叶病毒衣壳蛋白(TAT-TMVCP),与具有聚集诱导发光性质的有机铂金属大环(TPE-Pt-MC),通过静电相互作用自组装。在形成的组装体中,被包裹在内部的光敏剂TPE-Pt-MC提供活性氧产生能力,暴露在外表面的TAT提供穿膜能力,实现了穿膜增强的光动力抗菌。在光照条件下,该组装结构可明显抑制大肠杆菌及金黄色葡萄球菌的生长。尤其对于具有外膜结构的革兰氏阴性菌——大肠杆菌,该组装结构可使其存活率由黑暗条件下的~55%降低至光照条件下的~0%。该研究对于提高光动力抗菌效率,以及制备多功能纳米粒子具有重要意义。相关研究成果以Membrane Intercalation-Enhanced Photodynamic Inactivation of Bacteria by a Metallacycle and TAT-Decorated Virus Coat Protein为题,发表于PNAS。

中科院理化技术研究所牛忠伟课题组博士生高偲嘉为该论文的第一作者,上海交通大学颜徐州研究员、美国犹他大学Peter J. Stang教授、中科院理化技术研究所田野副研究员是该论文的通讯作者。
